Where NAD+ comes from
Cells obtain NAD+ by three routes. The salvage pathway recycles nicotinamide back into NAD+ and accounts for most day-to-day turnover. The de novo pathway builds it from tryptophan. The Preiss-Handler pathway starts from nicotinic acid.

Most product directions suggest morning, on the reasoning that NAD+ levels follow a circadian pattern. Published human evidence for a specific optimal time is limited; the honest answer is that consistency is better established than timing.
With or without food?
