Resveratrol Research: From Harvard Labs to the Supplement Aisle

Resveratrol became famous through work associated with David Sinclair's laboratory at Harvard Medical School in the 2000s, reporting that it activated SIRT1 and extended lifespan in model organisms. That research is also the reason the compound has been argued over ever since.

How resveratrol became a household word

Three threads converged. Epidemiological interest in the so-called French paradox pointed at red wine. Laboratory work reported lifespan extension in yeast, worms and flies. And in 2006, a study in mice on a high-fat diet reported improvements in metabolic markers and survival with resveratrol supplementation.

A biotech company, Sirtris, was founded on the sirtuin-activation thesis and acquired by GlaxoSmithKline in 2008 for around 720 million dollars. That transaction, more than any single paper, is why resveratrol became a consumer product category.

The scientific argument that followed

The original SIRT1 findings were challenged. Several groups reported that the activation observed depended on a fluorophore attached to the test substrate, and did not appear with native substrates. Later work proposed indirect mechanisms, including AMPK activation, rather than direct sirtuin binding.

Where this leaves a buyer. Resveratrol is one of the most studied polyphenols in existence, and its mechanism in humans is still argued over. Both of those statements are true simultaneously. Anyone presenting resveratrol as settled science, in either direction, is simplifying.

What the human trials look like

Human studies of resveratrol have been conducted, generally small and short, with mixed results and considerable variation in dose and formulation. The recurring complication is the one covered on our bioavailability page: free trans-resveratrol in plasma is very low after oral dosing, so trials differ enormously in what participants were actually exposed to.

Why we still make it

Because it is a well-characterised molecule with a large literature, it can be manufactured to a verifiable purity, and it can be tested for contaminants such as emodin. Those are things we can document on a certificate. The mechanistic debate belongs to researchers, and we would rather point you at it honestly than pretend it concluded.
